Continuous perfusion culture for optimization of process parameters
Octapharma Biopharmaceuticals develop processes for production of recombinant proteins. Process optimization is an important task which is often performed in 2 L bioreactors run in either batch or continuous perfusion mode. By using continuous mode it is possible to evaluate the effect of one process parameter while keeping other process parameters constant. One difficulty with continuous cultures is to keep the cell density constant for a long period of time. Presently the cell density is controlled by continuously bleeding off cells from the bioreactor. The bleed rate is controlled by a pump which is manually adjusted each day. Manual adjustment of the bleed pump often leads to deviations from the target cell density of the process. This is especially the case when growth rate changes after a process change. An optimized method for controlling cell density at set-point in the continuous culture is therefore desired. It has been suggested to use either a biomass monitor or an optical sensor to control the cell density more accurately.
The purpose of the project is to set-up a continuous culture run in perfusion mode where the cell density is controlled accurately at the set-point with a bleed pump controlled by a feed-back loop from either a biomass monitor or an optical sensor. Another purpose is to optimize process parameters with regard to productivity and product quality in the continuous culture.
Diploma work, 20 weeks.
The project encompasses common cell culture techniques, such as suspension cell culture in shaker flask and bioreactor. A basic literature search will be performed early in the project.
