Optimization of bioreactor cultivation protocol
Octapharma Biopharmaceuticals develop processes for production of recombinant proteins. One bioreactor model process use perfusion to achieve high cell densities. Cell specific perfusion rate (CSPR) is an important parameter to control in order to achieve a robust process with high productivity. The present process uses a step wise increase of the CSPR to compensate for the increased cell density during the time-course of the cultivation process. It is believed that a more smooth increase of the CSPR during the cultivation process would be beneficial with regard to productivity and product quality. A smooth increase could either be achieved by setting up a CSPR profile for the culture or by using a biomass monitor or optical sensor that adjust the CSPR with regard to the cell density in the process.
The purpose of the project is to optimize a bioreactor cultivation protocol in order to achieve a more robust process with regard to controlling CSPR at a desired set-point. Another purpose is to optimize other process parameters with regard to productivity and product quality in the cultivation model process.
Diploma work, 20 weeks.
The project encompasses common cell culture techniques, such as suspension cell culture in shaker flask and bioreactor. A basic literature search will be performed early in the project.
