Nitric oxide dependent mechanisms in the urogenital tract

Urologlab, Institutionen för molekylär medicin och kirurgi, Karolinska institutet, söker studenter som vill göra examensarbete (10-20 p). Möjligheter till senare doktorandprojekt finns. Lämplig bakgrund är högskolestudier i biomedicin, molekylärbiologi, cellbiologi eller biokemi.

Nitric oxide (NO) is an important biological messenger, involved in vasodilatation, inhibition of platelet aggregation, neurotransmission and host defence reactions. NO is a free radical, highly diffusible and hydrophobic, that rapidly converts in vivo into secondary reactive nitrogen species. High levels of NO are cytotoxic whereas low levels promote cell proliferation. Thus, it seems like NO is an important regulator of cell growth. NO is released in large quantities during inflammation and has been shown to have an important role in host defence mechanisms and may exert both anti-microbial and anti tumour activity. The aim of this project is to explore the role of NO in host defence reactions in the urogenital tract and to study the role of NO in human bladder and prostate cancer. We have seen increased levels of NO in bladder cancer patients with carcinoma in situ (CIS) and the production of NO increases dramatically in the bladder after bacillus Calmette-Guerin (BCG) treatment. The exact mechanisms behind BCG treatment are not known and approximately 30 per cent of patients that receive BCG do not respond to treatment. The project combines in vitro assays using urinary bladder cancer cell lines, in vivo assays using NOS knock out mice and patient tissues. The nitric oxide studies are well established in a strong research environment, with collaborations within Karolinska Institutet as well as internationally.

Candidates require excellent interpersonal skills and need to be able to work efficiently on their own initiative as well as in a team. Analytical thinking and effectiveness are essential characteristics. Practical experience in basic laboratory techniques e.g. aseptic tissue culture, immunoblotting, staining techniques is an advantage. Applicants should be able to communicate well in English as well as Swedish.
