Dioxin and light and skin cancer. Mechanisms for skin pigmentation at combined exposures to chemical carcinogens

Arsenic and polycyclic aromatic compounds such as benzo[a]pyrene (B[a]P) and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) are risk factors for skin cancer. Such compounds activate the melanogenesis and cause hyper pigmentation long before the tumours occur. This project is based on a new line of research, where we hypothesize that the aryl hydrocarbon receptor (AHR) activates transcription of melanocortin genes, as part of a stress response in the skin. Several AHR response elements have been identified in the transcription regulating sequence of the proopiomelanocortin (POMC) gene. We study the effects of low concentrations of inorganic and methylated arsenicals alone and together with B[a]P, TCDD or the tryptophan photoproduct 6-formylindolo[3,2-b]carbazole, in assays designed to determine activation of melanocortin gene expression, tyrosinase activity and cell proliferation.

The goal of this project is to clarify combined effects, at the molecular level, of the highly carcinogenic compounds arsenic and polycyclic aromatic compounds, which all cause skin effects in humans (pigmentation and hyperkeratinization).

Human epidermal keratinocytes and melanoma cells are used for measurements of gene transcription using reporter constructs and RT-PCR based methods.

The work is independent but will be performed together with other members of the research group. Knowledge about basic molecular techniques and some laboratory experience is necessary. A background in biochemistry, biology, biotechnics or medicine and an interest and curiosity about toxicological mechanisms is a good basis for this work. The exam work will be reported both orally and in a short written report.
