The effect of freeze-drying and granulation on the swelling properties of polymers

Polymers are normally the core component of most modified release formulations. Due to the dynamic structure of these materials, it is often hard to predict their confirmation and behavior in solution. When the functional groups of the polymers come in contact with their surrounding solution, they will slowly start to expand, to form a gel layer on their outer surface. However, It is believed that once the polymers have formed a network, the water is retained inside the structure and the entity starts to expand and swell. This phenomenon is of importance for the drug release profile from the unit.

Though some polymers are sensitive in terms of degradation, different processing methods are normally needed in their handling. Freeze drying is a commonly used drying technique for materials sensitive for undergoing changes during water elimination. However, limited information is available regarding further processing of this type of material in conventional pharmaceutical equipment. One way of facilitating the handling of not only freeze-dried material but most powder blends are by granulation.

The focus of this research is related to granular systems consisted of freeze-dried hydrophilic polymers.

Objectives
The scope of this work is to study granulation of freeze-dried polymers with respect to granular characteristics and swelling properties.

More specific the following will be studied:

* The characteristics of primary freeze dried particles and granules, in terms of size, morphology, flowability and compactibility.

* The influence of different process parameters during granulation, on the quality of the granules and their behavior in solution.
