Searching for Genes that Cause Type 2 Diabetes and Obesity

This project entails working with the latest in genomics technologies, particularly systems for genome variation analysis, to explore the genetic etiology underlying Type II Diabetes and Obesity.

Diabetes and Obesity are ´metabolic imbalances´ that have a large genetic component. A few genes or chromosomal regions are known that cause or contribute to these pathologies, but the primary etiologies are mostly unknown. Many more genetic risk components therefore remain to be discovered.
By using forefront molecular genetic techniques, we are searching for ´normal´ gene variations that are noticeably enriched in Type2 Diabetes and Obesity individuals, indicating a role in disease causation. The ultimate goal is treatment and prevention of these diseases.
Candidate genes we are testing include
i) components of glucose metabolism,
ii) genes for the transfer of insulin signals into the cell, and metabolic responses to it,
iii) genes previously suggested to be relevant to the disease processes (e.g., from animal models), and
iv) genes in chromosomal regions shown to harbor risk sequences by linkage analysis.
To study so many candidate genes we are using state-of-the-art robotic devices and large-scale polymorphism screening procedures. Bioinformatics and genome databases play a large part in this activity. The project will entail contributing to these research endeavors, thereby learning about the latest technologies, strategies, and problems associated with the analysis of complex disease genetics.
