Development and comparison of analytical methods for the quantification of residual host cell DNA

In the production of protein pharmaceuticals expressed in mammalian cells there is a need to remove DNA which is present as a process impurity from the cells. Often there are very small amounts of DNA present and in order to quantify those amounts there is a need for sensitive analytical methods. Depending on the stage in the production there are different requirements. In the process development stage there are a large number of samples and the need is for an analytical method with high throughput. In the final stages of the production there are not as many samples but there is a higher requirement on the sensitivity of the analytical method.
The goal of the project is to set up methods for the quantification of DNA and to evaluate and compare the methods and their applications. Potential techniques could be qPCR, spectroscopy, fluorescence or immunological methods.
The student will optimize the methods with DOE (Design Of Experiment) and evaluate and compare them with the existing method.
We would like the applicant to have good theoretical knowledge in relevant analytical methods and statistics and a genuine interest in problem solving and method development. Knowledge in chemometrics is a merit but not a requirement.
