Genetic engineering in cyanobacteria (Master thesis work at KTH 30hp)

The newly-formed Metabolic Engineering group at KTH Bioteknologi offers a Master thesis project for Spring 2012. The project theme is biofuel production from cyanobacteria. Recent studies have shown that photoautotrophic cyanobacteria can be used to produce biofuels using only carbon dioxide and light as food and energy sources. However, techniques for the manipulation of the cyanobacteria genome are not as refined as those for E. coli and yeast. The student will develop three platforms for the facile editing of the cyanobacteria genome, including mutation and deletion of native genes and the insertion of heterologous genes. The platforms will be used to optimize the expression of a five-enzyme biofuel pathway. The project will expound upon several key concepts in molecular biotechnology such as bacterial genetics, molecular cloning, cell-growth kinetics, and phenotype screening.
