Immunostaining-based molecular characterization of filopodia tips in neuroblastoma cells

Myosin-X has been found in our laboratory to bind the cytoplasmic tail of integrin beta subunits. Integrins are responsible for anchoring cells to the extracellular matrix. Myosin-X is a peculiar motor protein in that it localizes at the tip of filopodia, cellular extensions involved in adhesion and sensing of the environement. Myosin-X shuttles up and down filopodia, is involved in their elongation and is presumed to transport some molecules to their tip. However its role and mode of action are not well understood.

The aim of this study is to characterize the role Myosin-X plays in filopodia in relation to integrins and actin. Part of the approach taken in our laboratory is to label proteins present at filopodia tips in cells where Myosin-X has been altered by overexpression of the full-length or mutant protein or by siRNA-mediated knock down.

Ultimately the role of Myosin-X in axon guidance will be investigated. Therefore neuroblastoma cells are used for this study. Dermal fibroblasts (GD25) bearing beta1 integrins defective for Myosin-X binding are also used here.
