An evaluation of a new concept for UV detection likely to have a large impact on the way that we are working with LC

Today UV-detection is the dominating detection principle within the pharmaceutical industry. In combination with liquid chromatography (LC) it is the main technique for the determination of dose strength, impurities and degradation products, enantiomeric purity etc. One limitation associated with UV-detection is, however, that unstable and drifting base lines often are obtained as a result of mixing noise, ghost peaks, eluent/buffer absorbance, refractive index effects, temperature effects, etc. Another important limitation is that the UV-response may differ from substance to substance at a certain wavelength.
The purpose of the suggested diploma work would be to evaluate a new software that currently is under development. The algorithm, which is based on the deconvolution of diode-array data, has potential to offer the following advantages:
· Elimination of baselines effects ¿ ¿perfect¿ baselines
· Significant noise reduction ¿ lower detection limits
· Improved quality of UV spectra ¿ ¿error-free spectra¿
· A more general detection ¿ ¿concentration¿ chromatograms
· Infinite chromatographic resolution ¿ separation of co-eluting peaks
· Improved applicability for automated integration
This would result in a significant improvement of all LC performance parameters (sensitivity, linear dynamic range, selectivity, speed, accuracy, precision and robustness) and thereby dramatically change the way that we are working with LC.
